They can be wrapped once installed, but it is much, much, much easier to wrap them off the car.
You can run without heatwrap for awhile and then add it. But I don't reccomend ever removing it then. The heat wrap leaves behind funny marks that only professional polishing can take off.

And a lot of times header wrap voids the warranty (if any) on the headers. Some of the materials/coatings/welds are not designed to keep that much heat in (they are designed to radiate some and flow the rest out).
The major point of concern is where the wrapping ends, becuase that will be the first place the heat wants to radiate, and there will be a lot of it.
However, there are a lot of benefits to the wrapping, I just wanted to warn you of some of the consequences.
